Objectives

  • The aim of PRIM-TISS is to validate at laboratory scale TRL4:
  • a new product (a matrix system based on polylactic acid (PLA) and nano-hydroxyapatite (nano-HAP) with embedded silver (Ag) and Doxycycline(Doxy)
  • a new method of treatment (personalized multimodal and sequential treatment targeted against periodontal pathogens) of periodontal disease.

WP1. Study design of PRIM-TISS and preliminary results (TRL3) (8 months)

  • WP1.1. Review of existing technical documentation: therapy and intelligent matrices (nanofibers) (A2.1).
  • WP1.2. Development of nanofibers (nanofiber + active substance) to TRL3 and their physico-chemical characterization (A2.2+A2.3+A2.4)
  • WP1.3. Preparation and in vitro characterization of Doxy-based mucoadhesive topical drug delivery system able to be used in the in vivo studies for the topical noninvasive treatment of periodontitis (A2.4).
  • WP1.4. Material and method for obtaining the mucoadhesive system TRL4 - technology validated in lab (A2.4)
  • WP1.5. Results dissemination (D1+D2)

WP2. Therapy integration studies - personalized intelligent matrices and demonstration of the concept's functionality (TRL3) (12 months)

  • WP2.1. Nanofibers experimentation: adsorption/ desorption and kinetics studies (A2.4).
  • WP2.2. Establishment of sequential release of active subtances from nanofibers (A2.4)
  • WP2.3. Development of the antibiotics embedding in the mucoadhesive system with slow release (A2.3.).
  • WP2.4. Elaboration of a clinical model of implantation of the intelligent matrix in the patients (A2.2)
  • WP2.5. Realization of the personalized intelligent matrix by adsorption and incorporation of the antibiotic necessary and useful to each patient, according to the initial antibiogram (A2.4).
  • WP2.6. Application of personalized sequential treatment (A2.4) Data analysis and interpretation.
  • WP2.7. Clinical follow-up of patients for 3 months.
  • WP2.8. Demonstrating the utility and functionality of PRIM-TISS EM (A2.6)
